Abstract:对西门利克森林病毒(SFV)复制子质粒型表达载体pSfV1进行改造,即在SFV非结构蛋白(nSPs)上游插入依赖于RNA聚合酶Ⅱ的人巨细胞病毒立即早期启动子/增强子序列(hCMV IE Pro/Enh),同时在pS-FV1的多克隆位点下游插入SV40 poly(A)终止信号序列,获得可以完全在DNA水平上操作并具有自主复制功能的表达载体pSFV-DNA。为了进一步探讨改造载体的转染效率、表达能力以及是否能诱导细胞凋亡等特性,将报告基因EGFP插入pSFV-DNA中亚基因组启动子下游,构建的重组表达质粒pSFV-EGFP转染BHK-21细胞,转染后12h即可观察到荧光,但转染效率明显低于直接由hCMV IE Pro/Enh驱动的EGFP真核表达质粒pEGFP-C1;提取转染后48h的细胞基因组DNA,电泳发现pSFV-EGFP转染细胞均呈典型的DNA断裂(ladder),而pEGFP-C1转染细胞未观察到这种现象。上述结果表明:改造的载体不仅可完全在DNA水平上操作,而且能正确表达外源基因和诱导细胞凋亡,为开展各种病原微生物的“自杀性”DNA疫苗研究提供了良好的工具。

Abstract:将伪狂犬病毒Ea株纯化后免疫雌性Balb/c小鼠,取其脾细胞与SP2/O的骨髓瘤细胞融合,用间接酶联免疫吸附试验(ELISA)进行筛选,获得了2株抗PRV的单克隆抗体的杂交瘤细胞,分别命名为3G7E3和4A6F5,其细胞培养上清及小鼠腹水效价(ELISA)分别为1:212、1:212及1:100×212、1:100×212。特异性试验和免疫荧光结果显示,3G7E3和4A6F5仅与PRV反应,而与PPV、PRRSV、JEV和HCV等病毒不发生交叉反应;用单抗建立的夹心ELISA试验能检出伪狂犬病毒。表明本研究所获得的2株单抗是PRV特异性的。这2株分泌抗PRV单抗的杂交瘤细胞株的获得为进一步建立准确快速的抗原检测方法奠定了基础。

Abstract:分别用3种不同浓度的水杨酸(SA,3.25×10-5,7.5×10-5,1.5×10-4mol/L)处理水稻红莲型细胞质雄性不育(HL-CMS)型不育系粤泰A(YTA)和同核异质的保持系粤泰B(YTB)的三叶期幼苗,研究了幼苗叶片蛋白质含量及细胞壁纤维素和木质素含量的变化。结果表明,外源SA对水稻HL-CMS不育系和保持系的作用效果明显不同。3种浓度的SA均对YTA叶片蛋白质含量的增加起一定的促进作用,其中高浓度SA (1.5×10-4mol/L)的促进作用最为显著,但高浓度的SA对YTB则表现出抑制作用。与对照组(木村B培养液处理)相比,3.25×10-5mol/L和7.5×10-5mol/L的SA均能增加YTA三叶期幼苗叶片中细胞壁纤维素含量,而较高浓度(1.5×10-4mol/L)SA处理的作用不明显。对于YTB,只有较高浓度(1.5×10-4mol/L)的SA 对幼苗叶片细胞壁纤维素含量表现出明显的增加作用,3.25×10-5和7.5×10-5mol/L的SA处理没有明显的影响。YTA和YTB幼苗叶片细胞壁木质素含量的变化对SA的响应浓度有所不同,在较高浓度范围的SA能增加YTB幼苗叶片细胞壁木质素含量,而较高浓度范围的SA对YTA幼苗叶片细胞壁木质素则表现为抑制效应,SA仅在较低浓度范围有促进作用。同核异质系YTA和YTB间叶片蛋白质、纤维素和木质素含量的变化对外源SA处理的响应不同,可能只与YTA和YTB细胞质之间的差异有关。

Abstract:在实验室条件下对病原菌的形态特征、致病性和生物学特性进行了初步的研究。结果表明,引起草莓白绢病的病原物是齐整小核菌Sclerotium rol fsii Sacc.;病菌在偏酸性环境下生长较好,菌丝最适生长pH为5.08,在所测试的碳源中,S.rolfsii Sacc.对淀粉的利用最好,对乳糖利用效果最差;在供试氮源中,对硝酸钙和硝酸钠的利用最好,对尿素的利用最差;菌丝致死温度为50℃10min,菌核萌发的抑制温度为55℃10min。室内PDA平板药剂筛选结果表明,10%世高对病原菌菌丝生长的抑制效果最好。

Abstract:采用含毒介质法和牛津杯法测定了蛇床子素对8种植物病原真菌和7种病原细菌的抑制效果。结果表明:当浓度在400~800μg/mL时,蛇床子素对8种病原真菌都有不同程度的抑制作用,其中对水稻纹枯病菌和小麦赤霉病菌的抑制作用最佳,抑制率在95%~100%,其EC50分别为37.71μg/mL和47.06μg/mL,但对蕃茄叶霉病菌和玉米小斑病菌抑制作用较差;蛇床子素对7种病原细菌抑制作用最强的是水稻细菌性褐条病和甜豌豆带化病菌,浓度在1000μg/mL时,其抑菌圈直径分别为37mm和36mm,但对兰花褐腐病菌抑制力很低,只有8mm。

Abstract:在15~50℃、电机转速为75~750r/min下测定了苏云金芽胞杆菌发酵液及其微滤浓缩液的流变特性。分析结果显示:幂律方程适于描述发酵液及其微滤浓缩液的流变曲线,它们的流变指数n值均小于1(n=0.735~0.975),这表明苏云金芽胞杆菌发酵液及其微滤浓缩液均为拟塑性流体。随着浓度的上升,流体的稠度系数上升,流变指数下降;随着温度的上升,稠度系数下降,流变指数略有上升。经回归分析发现,温度对稠度系数的影响符合Arrhenius方程K=K0exp(Eu/RT),浓度与稠度系数的关系符合关系式K-aexp(bC^2),不同温度下的流变指数平均值nave和浓度的关系可表示:nave=1.35exp(-0.0067C)。

Abstract:研究了分离自仔猪肠道的乳酸菌L5和L7对低pH值、胆盐的耐受性。结果表明,pH值在2.5、3.5和4.5时,L5菌株处理150min对其无显著影响,在pH1.5时经30min处理,L5仍可维持较高的存活率;L7菌株在pH3.5和4.5条件下经150min处理后可保持较高的存活率,而在pH1.5和2.5条件下经处理30min后菌株数量显著下降。胆盐对L5和L7菌株的生长有一定影响,但低浓度短时间内影响较小。在同一胆盐浓度下,随培养时间的增长,活菌数下降。当胆盐浓度高于0.2%时,L5和L7菌株的生长极差。热稳定性试验结果表明,L5菌株在50℃下可成活,且呈正常生长趋势,但其对80℃及以上的耐热能力显著降低。

Abstract:采用放射免疫、RT-PCR等方法,研究了酪啡肽对21日龄断奶仔猪消化道酸度、血清中胃泌素水平和胃肠道胃泌素mRNA表达的影响。将24头仔公猪分对照组(正常哺乳28 d断奶)、试验I组(β-酪啡肽-7组) 和试验Ⅱ组(早期断奶组)。试验组于21 d断奶后,再连续添喂β-酪啡肽-7或生理盐水,每天2次(共20 mL)。10 d后,3组动物各随机取6头同时宰杀,共测定了18头仔猪胃、空肠和回肠食糜pH值及血清中胃泌素的含量,并对胃幽门和十二指肠胃泌素mRNA的表达进行了分析。结果发现日粮中添加β-酪啡肽-7的仔猪胃内平均pH为2.76±0.51,低于对照组(5.08±0.43)和早期断奶组(4.26±0.62);空肠和回肠食糜中pH值各组无明显差异。血清中胃泌素含量β-酪啡肽-7组最高,为100.03±18.72 pg/mL,与对照组相比差异极显著。RT- PCR检测分析表明:β-酪啡肽-7组仔猪胃窦胃泌素mRNA表达量高于对照组(P<0.01)和早期断奶组。十二指肠中胃泌素mRNA表达的变化情况与胃窦相似。认为酪啡肽类可以促进断奶仔猪的胃酸分泌,其机制可能是通过上调胃泌素mRNA的基因表达,使胃泌素分泌增加所至。

Abstract:选100羽8周龄健康海兰褐商品蛋鸡,通过关节腔直接注射金葡菌的方法复制鸡葡萄球菌性关节炎的病理模型,并在饲料中添加一定比例的中药进行治疗,运用血清学方法对试验动物的血清生化指标进行动态研究,从而探讨中药方剂对鸡葡萄球菌性关节炎的治疗效果。研究结果发现:金荞麦、艾叶等中药组成的方剂可以明显地恢复血清中与肝脏、心脏和肾脏功能相关的生化指标如白蛋白(Alb)、总蛋白(TP)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH-L)、α-羟丁酸脱氢酶(α-HBDH)和尿酸(UA)的水平,从而在鸡葡萄球菌性关节炎发生发展中可以保护以上各个脏器免受损伤。因此认为,金荞麦、艾叶等中药组成的方剂可以预防和治疗鸡葡萄球菌性关节炎。

Abstract:研究了采后冷藏、外源ABA、亚精胺(Spd)、1-甲基环丙烯(1-MCP)、热处理及酸处理对荔枝果实脂氧合酶(LOX)活性变化的影响。荔枝果实组织中LOX活性测定反应系统的最适pH是6.5。与3℃冷藏相比,0℃下糯米糍果皮LOX活性升高较快,丙二醛(MDA)含量积累加快,在21d时分别高出34.9%和25.6%。0℃下桂味果皮LOX活性先升后降,于14d达到高峰。冷害促进了果皮LOX活性增加,但0℃下糯米糍果肉的LOX活性和MDA含量均低于3℃下果肉。外源ABA、Spd、1-MCP处理不同程度地降低了果皮LOX活性,减轻了果实冷害的发生程度。推测由LOX主导的膜脂过氧化作用引起的膜结构破坏在荔枝果实衰老和冷害褐变中可能起重要作用。46.2~80.0℃的热水处理诱导LOX活性的升高,适当浓度的酸则显著抑制该酶的活性。

Abstract:用耐氨固氮菌浸种处理黄瓜种子,研究了低温胁迫下黄瓜幼苗保护酶活性的变化。结果表明:5℃低温胁迫2 d,清水组黄瓜幼苗叶片相对电导度、MDA含量及CAT活性显著升高,SOD及POD活性、叶绿素含量显著降低,恢复1 d,这种差异更显著,活性变化不明显;经耐氨固氮菌浸种处理后,菌液组黄瓜叶片叶绿素含量及保护酶活性显著增强,5℃低温胁迫2 d,也表现出清水组的变化规律,但CAT活性显著降低,恢复1 d,菌液组黄瓜叶片相对电导度、MDA含量则显著低于清水组(P<0.05),叶绿素含量则显著高于清水组(P<0.05), SOD及CAT活性显著升高(P<0.05),POD和AsA-POD活性则降低。结果显示,经耐氨固氮菌浸种处理后, 黄瓜幼苗提高了机体的抗氧化酶活性而增强耐寒能力,避免低温的伤害。
