Abstract:以杂交中稻红莲优6号为材料,采用小区试验,研究了不同施氮水平和栽插密度及栽插模式对杂交中稻群体结构的影响。结果表明:杂交中稻红莲优6号在适宜的密度(2.7×105穴/hm2)和施氮水平(纯氮210 kg/hm2)下,采用宽行窄株的栽插方式,可以增加群体叶面积指数,提高光合速率,改善群体光环境,从而达到增加产量的目的。

Abstract:为了探讨半夏试管珠芽形成的生理生化机制,以半夏无菌叶柄为材料,倒置在MS基本培养基上,对半夏珠芽形成过程中5种内源激素(IAA、GA3、ABA、ZR、JA)的动态变化进行了研究。结果表明:倒置半夏叶柄的珠芽无一例外地出现在叶柄形态学上端;ELISA方法测定显示,半夏叶柄形成珠芽的过程中叶柄上端IAA、ABA、ZR和JA的含量呈上升趋势,GA3在的珠芽形成初期急剧下降。随着珠芽的形成,GA3与其它各激素的平衡关系发生变化,IAA/GA3、ABA/GA3、ZR/GA3和JA/GA3的比值先呈下降趋势,15 d后开始上升;在珠芽快速形成期(10~20 d)IAA/GA3、ABA/GA3和JA/GA3维持在高的水平,其中JA/GA3的比值20 d后又呈上升趋势。

Abstract:对标准的链霉菌接合转移方法进行优化建立起适用于吸水链霉菌NRRL5491的接合转移系统。40℃温浴20 min为孢子的最佳热激处理条件;整合型质粒pSET152的接合转移效率是穿梭型质粒pOJ446的3~6倍;当阿泊拉霉素覆盖浓度为1μg/mL时,pSET152的接合转移效率是50μg/mL时的4.6倍,而pOJ446接合转移效率相应提高了2.4倍;MS培养基上接合转移效率是SY琼脂培养基上的3.4倍。采用最适优化条件时,接合转移频率可达到2×10-6,足够用于基因置换等遗传操作。

Abstract:原生质体制备与融合是重要的细胞工程技术,有利于实现远缘遗传物质的直接交换,促进遗传资源创新。本试验用Olympus BHS显微镜观察了曲霉原生质体融合全过程;采用淀粉-麸皮-硫铵培养基、淀粉-麸皮-豆粕培养基、马铃薯蔗糖琼脂培养基(PDA)等研究曲霉融合子及亲株产淀粉酶能力的特点;比较分析了亲株和融合子产淀粉酶性能与温度稳定性;比较研究了曲霉亲株和融合子淀粉酶的淀粉-聚丙烯酰胺非变性垂直平板凝胶电泳特征和各菌株可溶性蛋白质的淀粉-聚丙烯酰胺凝胶电泳特征。结果表明:曲霉原生质体融合过程很快,整个融合过程在5 min之内完成;融合子及亲株产淀粉酶随培养基、培养时间不同而变化;F05融合子在淀粉-麸皮-豆粕培养基产酶水平达(61 111.42±123.30)U/g,比亲株M70和M4分别高204.81%和114.00%,耐高温性能也有所提高;2种电泳结果都能够特异性显示淀粉酶带、重复性好。可见,原生质体融合是改良曲霉淀粉酶的有效方法。

Abstract:采用体外法研究了不同精粗比底物下(A组为全粗料,B组3∶7,C组5∶5,D组7∶3,E组为全精料)瘤胃真菌发酵特性及其对粗饲料(稻草秸)的附着情况。发酵试验结果表明:随底物精料比例的增加,同一时间点真菌数量呈上升趋势,A组到E组发酵96 h的累计产气量显著上升,分别为177.2 mL、198.2 mL、217.4 mL2、33.4 mL和251.5 mL,提示底物中精料比例的增加能促进瘤胃真菌的生长和发酵。此外,随底物精料比例的增加,羧甲基纤维素酶和α-淀粉酶酶活呈上升趋势,木聚糖酶酶活则呈下降趋势。稻草秸附着结果表明:不同发酵时期底物精粗比对附着于稻草秸的瘤胃真菌数量有很大影响。发酵前期(24 h),精料组(B组、C组和D组)附着于稻草秸表面的真菌孢子囊数显著高于粗料组(A组)。发酵后期(72 h),A组和B组附着于稻草秸表面和切缘的孢子囊数高于C组和D组,A组和B组附着于稻草秸表面和切缘的孢子囊数与48 h相比无显著变化,而C组和D组附着于稻草秸表面和切缘的孢子囊数显著下降。这种附着差异可能与不同精粗比底物提供的能量和附着位点不同有关。

Abstract:为摸索适宜椰子的SSR反应体系,分析了PCR反应体系中的Mg2 浓度、引物浓度、dNTP浓度、TaqDNA聚合酶浓度、模板浓度以及退火温度对扩增结果的影响,建立了适合椰子的SSR反应体系。研究结果表明:在20μL反应体系中,Mg2 、引物和dNTP的最适浓度分别为2.5 mmol/L、0.3μmol/L、0.2 mmol/L;TaqDNA聚合酶的最佳使用量为1 U,模板DNA应加入50 ng,引物最佳退火温度比Tm值较小者低2~3℃。在参数优化的基础上,应用标记分析该反应体系对24个海南不同地区高种椰子样品进行SSR分析,不同样品间DNA谱带多态性丰富,本研究建立的分析体系将为今后椰子资源的SSR分析奠定良好的研究基础。

Abstract:利用包含双元载体pBI121的农杆菌LBA4404转化西瓜子叶外植体,获得转基因西瓜植株。结果表明:转化受体材料西瓜品种早花、克伦生、黑美人母本F23的分化效率没有明显差别;外植体浸染菌液10 min的处理较为合适;125 mg/L卡那霉素和300 mg/L羧苄青霉素适合于外植体筛选培养。通过PCR检测获得了3株转基因植株,转化效率为1.5%,其中只有2株整合了gus报告基因。Southern杂交和gus染色检测证明2株的基因组中整合了外源基因,报告基因在植株T0-1-3后代呈现3∶1分离。

Abstract:以白鲢鱼骨蛋白为对象,在确定复合蛋白酶(protamex)适宜水解条件的基础上,采用正交试验对复合蛋白酶与风味蛋白酶(flavorzyme)分步水解白鲢鱼骨蛋白的工艺进行优化。结果表明,复合蛋白酶的适宜反应温度为50℃,适宜反应pH值为6.5,最适加酶量为3 000 U/g。采用复合蛋白酶和风味蛋白酶分步水解鱼骨蛋白可以提高水解度和氮收率,最适水解条件是在料液比1∶4(W/V),起始pH 6.5,用1 500 U/g复合蛋白酶于50℃下水解2 h后,再用1 500 U/g风味蛋白酶继续水解2 h,其水解度和氮收率分别为28.75%和79.69%,比单独用复合蛋白酶或风味蛋白酶水解时,水解度分别提高11.07%和9.81%,氮收率分别提高5.17%和13.22%。

Abstract:以白鲢鱼糜为原料,在确定凝胶劣化温度的基础上,研究了食盐、水分和可溶性蛋白对凝胶劣化的影响。结果表明,在供试温度范围内(40℃、50℃、60℃和70℃),随着加热时间的延长,鱼糜凝胶的破断强度呈现先增加后下降的趋势,且加热温度越高破断强度达到最大值所需的时间越短、下降也越显著。鱼糜凝胶劣化温度段为50~60℃,凝胶劣化与该温度段肌球蛋白重链的降解密切相关;盐溶蛋白和水溶蛋白都能不同程度地抑制凝胶劣化,但超过一定添加量时就会产生相反的作用;凝胶劣化程度随含水量的增加而增大,也随食盐含量(1%~3%)的增大而增大。
