Abstract:以豆科植物百脉根为材料,构建了AD-cDNA表达文库。其库容量达到每3μg DNA 2×106酵母转化子,空载率为12.5%,平均片段长度为1.5 kb。以百脉根结瘤因子受体激酶基因1(nod factor receptor kinase,NFR1)蛋白激酶结构域的DNA片段(NFR1-PK)为诱饵,利用酵母双杂技术,筛选与NFR1相互作用的基因。结果在含X-gal的四缺选择性培养基上筛选到120个克隆。经质粒抽取、PCR鉴定、回转酵母验证得到80个阳性克隆。对26个确定阳性克隆的外源片段进行测序和同源性分析,通过NCBI数据库比对分析鉴定含有JAB1、AT-RICH结构域等4个与NFR1-PK相互作用的不同基因。通过半定量RT-PCR观察在百脉根中这些基因的表达情况,其结果可为进一步研究NFR1基因的功能和调控机制提供材料。

Abstract:为了提高酿酒酵母中β-葡聚糖(SCG)含量,本研究对酿酒酵母(Saccharomyces cerevisiaeFFL01)培养基进行了优化。通过单因素试验评价了碳源、氮源、无机离子、酶激活剂等对酵母生物量及SCG的影响,运用二次回归旋转组合建立数学模型,通过响应面分析确定培养基各主要组分最佳含量为:葡萄糖31.1 g/L、蛋白胨19.1 g/L、酵母膏15.0 g/L、甘油9.7 g/L。优化培养基使SCG由原来的(0.664±0.013)g/L增加到(1.112±0.008)g/L。所建模型二次项显著水平P<0.01,证明培养基对SCG影响达极显著水平。

Abstract:草甘膦N-乙酰转移酶(glyphosate N-acetytransferase,GAT)是一种能够通过转乙酰基的反应将草甘膦代谢为低毒物质乙酰草甘膦的酶,在抗除草剂转基因作物中具有潜在的应用价值。通过将地衣芽孢杆菌Ba-cillus licheniformis(Weigmann)Chester AB 94036中的草甘膦N-乙酰转移酶(glyphosate N-acetyltransferase,GAT)基因克隆到大肠杆菌中,获得含有重组质粒pGEX 6p-1-gat的重组菌株。对重组菌株进行诱导表达,经过GST亲和层析纯化,获得约17 ku的目的蛋白。通过酶标仪和连续分光光度分析法,对GAT蛋白进行了酶反应动力学测定。研究表明GAT的催化常数kcat为6.420 min-1,米氏常数KM为1.162 mmol/L,由此得出其催化效率kcat/KM为5.52 L/(mmol.min)。

Abstract:采用PCR-RFLP(MspⅠ)方法分析了湖北白猪杂交后代共计228头猪的MyoG基因外显子2、3及内含子2和5′-端调控区域遗传多态性,利用最小二乘模型分析了杂交后代MyoG基因型对相关生产性能的影响。结果表明:在PCR1MspⅠ-RFLP位点上,杂交后代的优势基因型为AA型,没有发现纯合BB型;在PCR2MspⅠ-RFLP位点上,杂交后代表现为单态,没有发现梅山猪特异性MspⅠ多态性酶切位点;不同基因型间个体初生重、20日龄重、日增重、瘦肉率、肌内脂肪含量等生产性能差异不显著。

Abstract:300日龄健康溆浦鹅种鹅300羽,随机分成5组,每组3个重复,每个重复20羽(公母比例1∶4),每间隔10 d分别注射不同剂量LHRH-A3(0、5、10、15、20μg/羽),研究其对溆浦鹅种鹅繁殖的影响。结果表明:注射10μg/羽和15μg/羽LHRH-A3组产蛋率、血清雌二醇浓度显著提高,料蛋比显著降低,10μg/羽组血清孕酮浓度显著升高;20μg/羽组产蛋率及血清中孕酮和雌二醇的浓度均显著降低,蛋重、蛋形指数和料蛋比显著升高。

Abstract:从患典型出血性败血症死亡的施氏鲟(Acipenser schrenckiiBrandt)脾脏组织分离到1株致病性病原菌。依Koch法则建立室内感染模型,一次感染试验和回归感染试验的试验组均可复制出与自然发病情况下相同的症状,根据Reed-Muench法计算病原菌半数致死浓度为1.17×107cfu/mL。对从自然发病和一次感染试验的病鱼体内分离的致病菌进行形态和VITEK全自动细菌生理生化表型鉴定以及16S rRNA基因部分序列进化树分析,结果表明导致施氏鲟出血性败血症的病原菌为嗜水气单胞菌(Aeromonas hydrophila)。药物敏感试验结果显示,该病原菌对庆大霉素、环丙沙星、阿米卡星等药物敏感,对氨卞青霉素、舒巴坦、头孢唑啉等药物具有抗性。

Abstract:以湖北省宜都市的斑点叉尾鮰为研究对象,利用8个微卫星位点IpCG0033、IpCG0035、IpCG0040、IpCG0041、IpCG0043、IpCG0051、IpCG0054、IpCG0057的引物进行了遗传分析,统计分析了等位基因数、杂合度、平均多态信息含量等遗传参数,并检测了群体符合Hardy-Weinberg平衡的状况。结果显示:8个微卫星标记位点在群体中共检测到47个等位基因,观测等位基因数在3~8个,平均有效等位基因数为4.254 8个,群体观测杂合度为0.787 3,期望杂合度为0.728 2,平均多态信息含量(PIC)为0.692 0,达到高度多态(PIC>0.5),8个位点中有4个(IpCG0033、IpCG0041、IpCG0054、IpCG0057)处于Hardy-Weinberg平衡状态(P>0.05),另外4个处于不平衡状态(P<0.05)。结果表明我国湖北省宜都市养殖的斑点叉尾鮰目前仍具有较高的遗传多样性,遗传信息丰富,遗传变异大,可以作为良好的育种材料。

Abstract:以鳃、肌肉和内脏组织超氧化物歧化酶SOD、过氧化氢酶CAT、谷胱甘肽过氧化物酶GSH-PX活性为指标研究了Cu2 污染对牙鲆的损伤作用。通过室内暴露试验,将牙鲆置于不同浓度(0.00、0.01、0.05、0.30、0.50、0.80 mg/L)的Cu2 溶液中10 d,测定其鳃、肌肉和内脏中过氧化氢酶CAT、谷胱甘肽过氧化物酶GSH-PX、超氧化物歧化酶SOD的活性,结果表明:随着Cu2 浓度的增高,牙鲆的3种组织中CAT、SOD、GSH-PX的活性均表现为:Cu2 浓度在0.01~0.05 mg/L出现不同程度的诱导,酶活性均高于对照组水平,Cu2 浓度大于0.05 mg/L时,各酶活性低于对照组水平。可以证实,SOD、CAT、GSH-PX活性变化可以反映牙鲆的伤害程度。

Abstract:研究了温度、pH、促进剂、抑制剂、菌体及免疫多糖对锯缘青蟹(Scylla serrata)血细胞裂解物(HLS)酚氧化酶(phenoloxidase,PO)活力的影响。试验结果表明:锯缘青蟹PO的最适作用温度为25℃,最适pH值为7.8;SDS、Ca2 、Mg2 均可提高PO的活力,其中Ca2 和Mg2 的最佳反应浓度分别为50 mmol/L和150 mmol/L,SDS的最佳反应浓度为2.5 mg/mL;叠氮钠、二乙基二硫代氨基甲酸钠、乙二胺四乙酸、Cu2 对PO活力有不同的抑制作用。发现拟态弧菌SS060818、气单胞菌Mr M0602、金黄色葡萄球菌ATCC25923、溶壁微球菌AS1.634对PO有激活作用,其中拟态弧菌和气单胞菌激活作用显著,金黄色葡萄球菌、溶壁微球菌对PO有弱激活作用;肽聚糖、拟态弧菌脂多糖粗提物可显著提高PO活力,肽聚糖、脂多糖的最佳激活浓度分别为2.7mg/mL、0.5 mg/mL。

Abstract:植物硫化激动素(phytosulfokine-α,PSK-α)是植物体内一种小分子肽类生长调节因子,具有促进细胞增殖等多种生物活性。但由于其在植物体内的含量极低,难以满足PSK-α研究和应用的需要。笔者在前期构建PSK-α原核型表达载体的基础上,研究了影响融合蛋白表达的因素,优化了培养条件,并运用谷胱甘肽S-转移酶(glutathione S-transferase,GST)亲和柱层析对融合蛋白进行了分离纯化。通过单因素和正交试验的结果表明:培养温度、诱导剂浓度、诱导剂加入时机和诱导时间均对融合蛋白的表达量和表达形式有很大影响;在诱导温度20℃,扩大培养150 min(菌液培养至OD600值约0.8止)后加入诱导剂1.0 mmol/L异丙基-β-D-硫代半乳糖苷(isopropy-β-D-thiogalactoside,IPTG),诱导培养时间8 h条件下,可以获得最多的可溶性融合蛋白GST-PSK-α表达量。

Abstract:以酵母-β1,3-葡聚糖酶解产物为原料,经超滤得到水溶性葡聚糖WSGI(water soluble-β1,3-glucanI,WSGI)(Mr>10 ku)和WSGII(Mr<10 ku),进行体内抗肿瘤试验,比较不同给药方式以及水溶性葡聚糖与羧甲基葡聚糖对肿瘤小鼠抑瘤率、胸腺指数、脾脏指数的影响,并测定脂质过氧化物丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力等抗氧化指标。结果表明,WSGI能明显抑制肿瘤S180的生长,抑制率可高达49.84%,并且呈现出剂量-效应关系,其抑瘤效果强于同剂量下羧甲基葡聚糖,与环磷酰胺阳性对照组相比差异不显著,腹腔注射给药抑瘤效果好于灌胃给药;同时WSGⅠ可显著增加小鼠体内的抗氧化活性;而分子量较小的WSGII则没有明显的抑瘤效果。

Abstract:以河南、湖南、云南、贵州、吉林与津巴布韦烤烟烟叶为材料,采用美国HP5890Ⅱ-5972气质联用仪定性、内标定量法对国内不同香气风格烤烟与津巴布韦烤烟烟叶中中性挥发性香气物质组成和含量进行了对比分析。结果表明:在所检测出的29种中性挥发性致香物质中,津巴布韦烤烟烟叶中12种香气物质含量最高,其中11种明显高于国内烟叶,如3,4-二甲基-2,5-呋喃二酮、2-乙酰基吡咯、巨豆三烯酮(A、B、C、D)、2-壬烯醛、螺岩兰草酮等,其烤烟烟叶质量和风格表现出显著的焦甜香气息,香气浓郁。国内河南、湖南等浓香风格烤烟内茄酮、糠醛、糠醇、乙酰基呋喃、2-乙酰基吡咯等西柏烷类降解产物和棕色化反应产物含量较高;云南等清香型烤烟中β-大马酮、二氢猕猴桃内酯和3-羟基-β-二氢大马酮等类胡萝卜素类降解产物含量较高。

Abstract:土壤中各种形态的钾之间存在着动态的平衡,这些平衡受一定因素的影响。本文主要从粘土矿物类型、土壤水分状况、温度、pH、肥料施用以及陪伴离子等方面对土壤钾素转化的影响因素进行了综述,可以归纳为:①以2∶1型粘土矿物为主的土壤吸附K 的结合能较小,易解吸;而以1∶1型粘土矿物为主的土壤吸附K 的结合能较大,吸附的K 不易解吸。②土壤干湿变化对钾素转化的影响因速效钾含量的高低而有明显的区别。③在0~40℃温度范围内,增温能使交换性钾离子减少,而水溶性钾离子浓度则相应增加。④pH对土壤钾素转化的影响比较复杂。⑤氮肥中的NH4 因与K 离子具有相似的离子半径和水化能而对土壤钾素的有效性产生显著的影响。最后展望了钾素转化领域未来的发展方向和研究热点。
